چکیده

Tissue preparation The segments of the right coronary arteries (2-3 cm from the origin) were excised from the porcine hearts at a local slaughterhouse immediately after the animals had been slaughtered. The segments were brought back to the laboratory in ice-cold normal physiological salt solution (PSS). After removing the adventitia, the segments were cut into strips in a circular direction, and then were opened longitudinally (approximately 1 mm wide, 5 mm long, and 0.1 mm thick). Care was taken to avoid damaging the endothelial cells. The strips without endothelium were prepared by rubbing off the inner surface with a cotton swab.

Plasmin induces endothelium-dependent nitric oxide-mediated relaxation in the porcine coronary artery.

OBJECTIVE Plasmin is a key enzyme in fibrinolysis. We attempted to determine the possible role of plasmin in the regulation of vascular tone, while also investigating the mechanism of plasmin-induced vasorelaxation. METHODS AND RESULTS In porcine coronary artery, plasmin induced an endothelium-dependent relaxation. This relaxing effect was mostly abolished by a proteinase inhibitor, a plasmin...

The Possible Involvement of Nitric Oxide/Endothelium Derived Relaxing Factor in Atropine-Induced Vasorelaxation

Atropine has been used to block cholinergic neurotransmission in basic research. Large doses of atropine cause vasodilation of the blood vessels in the skin. This effect is apparently unconnected with the antimuscarinic activity of atropine and seems to be due to a direct action on the blood vessels. It has been suggested that atropine blocks muscarinic receptors at low doses and it induces th...

Red Ginseng Saponin Fraction A Isolated from Korean Red Ginseng by Ultrafiltration on the Porcine Coronary Artery

Red ginseng saponin fraction-A (RGSF-A) contains a high percentage of panaxadiol saponins that were isolated from Korean red ginseng by ultrafiltration. The aim of this study was to elucidate the effects of RGSF-A on the porcine distal left anterior descending (LAD) coronary artery. The relaxant responses to RGSF-A were examined during contractions induced by 100 nM U46619 (9,11-dideoxy-9a,11a-...

Endothelium-mediated relaxation of porcine collateral-dependent arterioles is improved by exercise training.

BACKGROUND Endothelium-dependent modulation of coronary tone is impaired in the collateral-dependent coronary microcirculation. We used a porcine model of chronic coronary occlusion and collateral development to evaluate the hypothesis that exercise training enhances endothelium-mediated relaxation and increases endothelial nitric oxide synthase (ecNOS) mRNA levels of collateral-dependent micro...

Beta-adrenoceptor-mediated responsiveness of human internal mammary artery

The internal mammary artery (IMA) is currently the preferred conduit for myocardial revascularization. However, pre-operative vasospasm and a hypoperfusion state during maximal exercise may limit its use as a bypass graft. The mechanism of spasm has not been clearly defined. Since β-adrenoceptor activation plays a major role in vasorelaxation, the present study was carried out to investigate th...
